First New Gram-Negative Antibiotic Class Approved Since 2019
The US Food and Drug Administration has granted full approval to a novel siderophore cephalosporin antibiotic for hospital-acquired and ventilator-associated pneumonia caused by carbapenem-resistant Acinetobacter baumannii (CRAB) — one of the pathogens the WHO designates as a "critical priority" for new drug development. A Phase 3 trial published in The New England Journal of Medicine found the agent reduced 28-day all-cause mortality by nine percentage points compared with best available therapy (colistin plus sulbactam) in patients with culture-confirmed CRAB infection. This is the first new antibiotic class targeting Gram-negative drug-resistant bacteria approved by the FDA since 2019. The Infectious Diseases Society of America welcomed the decision but warned that pipeline investment for Gram-negative pathogens remains "critically underfunded." Pricing and reimbursement pathways will determine how quickly the drug reaches intensive-care units in middle- and lower-income countries, where CRAB mortality is highest.
Long COVID Brain Fog Traced to Persistent Neuroinflammation
A cohort study published in Nature Medicine followed 3,100 adults with confirmed long COVID over 18 months, combining PET imaging, cerebrospinal fluid analysis, and serial cognitive testing. Researchers within the NIH-funded RECOVER programme found that participants with persistent cognitive symptoms had significantly elevated markers of microglial activation and IL-6-pathway signalling compared with recovered controls — a pattern that did not resolve spontaneously in most cases. Brain fog severity correlated directly with the degree of neuroinflammation on PET imaging, providing the strongest mechanistic evidence to date that long COVID's cognitive effects have a measurable biological substrate rather than representing purely psychological sequelae. The study's authors stressed that this is observational data: IL-6 pathway inhibitors such as tocilizumab have not been tested in a controlled trial for this condition, and off-label use is not supported by current evidence. A randomised phase is now being planned.
West Nile Virus Peaks Across Greece and Southern Italy; Cyprus Raises Alert
The European Centre for Disease Prevention and Control reported this week that confirmed West Nile virus (WNV) human cases across southern Europe have reached the highest August count in a decade. Greece logged 84 confirmed cases and 11 deaths since May; southern Italy recorded a further 47 cases. Cyprus's Ministry of Health elevated its public advisory from "watch" to "alert" on Friday, citing increased mosquito surveillance data in the Mesaoria plain and Limassol coastal zone. No human cases had been confirmed on the island at the time of publication. Health authorities are urging residents to apply DEET-based repellents, eliminate standing water on their properties, and report dead birds, which serve as an early sentinel for viral circulation in a region. Persons over 65 and those who are immunocompromised face the highest risk of neuroinvasive disease; WNV has no approved antiviral treatment, and care is supportive.
Two-Year GLP-1 Trial in Adolescents: Semaglutide Cuts BMI by 16% Without Affecting Growth
A multinational two-year trial of once-weekly semaglutide 2.4 mg in 820 adolescents aged 12–17 with obesity, published in The New England Journal of Medicine, found a mean body-mass index reduction of 16.1% in the treatment arm versus 0.6% in the placebo group. Bone-age assessments and standing-height measurements at 24 months showed no significant difference between arms, addressing the primary safety concern that had slowed paediatric regulatory discussions. Gastrointestinal side-effects were common — nausea in 62% of treated participants, vomiting in 41% — but discontinuation rates were similar between groups. The European Medicines Agency has confirmed it is reviewing a marketing-authorisation application for the paediatric indication, with a decision expected in the fourth quarter of 2026. Clinicians should note that the trial studied semaglutide alongside intensive lifestyle counselling; the individual contributions of each component were not separately assessed.
PRIMA Trial: Mediterranean Diet Cuts Colorectal Cancer Risk by 22% Over Seven Years
The PRIMA study — a randomised controlled trial conducted across eight EU countries including Cyprus — followed 12,400 adults aged 50–70 at elevated colorectal cancer risk over seven years. Participants randomly assigned to an intensive Mediterranean dietary pattern (emphasising olive oil, legumes, vegetables, and oily fish, with red meat limited to twice weekly) had a 22% lower incidence of confirmed colorectal cancer compared with a control arm receiving standard dietary advice. The greatest benefit was seen in participants who maintained high adherence across the full study period. A mechanistic sub-study published simultaneously in Gut identified higher faecal butyrate production, lower mucosal IL-1β, and a more diverse gut microbiome in the adherent group — pointing to an anti-inflammatory mucosal environment as the plausible biological pathway. This is a primary-prevention RCT, making it one of the highest-quality pieces of evidence for dietary intervention in cancer risk to date, and consistent with — but methodologically stronger than — prior observational meta-analyses.
18-Month Data Confirm Maternal RSV Vaccine Protects Infants Through First Winter
Updated follow-up data from the MATISSE trial confirm that a single dose of Pfizer's ABRYSVO maternal RSV vaccine, given between 24 and 36 weeks of gestation, maintains 51% efficacy against lower-respiratory-tract RSV disease in infants through 18 months of life. Protection was highest in the first six months after birth (72% efficacy) and declined gradually thereafter, consistent with the expected trajectory of maternally transferred antibodies. The European Medicines Agency is expected to review this data in the context of a label update before the 2026–27 winter respiratory season. Ireland and Germany have already incorporated maternal RSV vaccination into their national immunisation programmes. Cyprus's Ministry of Health has indicated it is reviewing the MATISSE follow-up data ahead of a potential programme launch in 2027.
